And what did you clean the screen with? Typically, diesel fuel works best with a soft bristle tooth brush. Other chemicals 'could' melt the plastic olefin mesh and prevent the fluid from flowing. Thus, the filter is now trashed.

The olefin mesh flows only in one direction. It's tiny little pores like this under 10X magnification.

Actually I cleaned it with soap and water, I can hold it up to the light and see through it.

Well, not 100% sure, when I hold it up I can see through it an do not see any blockage or residue.

Can I remove this screen without draining the fluid?

I doubt the screen is your problem, however, just looking at the parts drawing it would look like your screen is up pretty high on the transmission case. If it is above the dipstick level then yes, you could pull, it, if below, you could not without loosing your fluid or at least a portion of it. I would lean toward figuring out how to get some fluid in the suction line going to the pump. I agree with bmavericik you are likely air locked.

Is there a slope near you. Have the front end dip down. Try burping out the air. Does the machine have a loader too? Try the top most bleed fittings too.
